Abstract (Abstrak)
Although green algae (Divisio Chlorophyta) only occupy <1% of global seaweed biomass production, from the cultivation sector, green algae show a trend of increasing productivity and diversification due to their bioprospecting in the food sector. In addition, green macroalgae also show a unique diversity of cytomorphology, ecophysiological properties, propagation capacity, and content of bioactive compounds compared to red macroalgae (Divisio Rhodophyta) and brown macroalgae (Classis Phaeophyceae). The characteristics of macroalgae are very plastic so conventional type identification (morphology) must be combined with molecular-based identification for correct classification. This study took samples of green macroalgae from the seaweed cultivation area of Puntondo, Takalar Regency. The purpose of this study was to determine and compare the extraction methods that gave the best results to the green macroalgae Caulerpa and Codium. Two extraction methods were tested, namely the conventional method (CTAB) and the KIT method using the DNAeasy Plant Mini Kit (Qiagen, Germany). The success of genomic DNA extraction was analyzed from the results of the quantity and quality of genomic DNA as well as the success of amplification using the extraction product as a template. The quantity and quality of genomic DNA were analyzed using the Qubit dsDNA HS (High Sensitivity) Kit on the Qubit 3 Fluorometer and the absorbance value of A260/280 using a spectrophotometer while the amplification success parameters were analyzed by looking the presence or absence of DNA bands. The results showed that the CTAB method gave a better concentration and absorbance ratio than the Kit method. In addition, the use of primers OPA-02, OPD-03, and OPC-11 only succeeded in amplifying Caulerpa lentillifera genomic DNA.
