Abstract (Abstrak)

The aims of this study is to compare thy coqcentration of exosome and the characteristic of p53 inside exosome before or after chemotherapy in patient with nasopharyngeal cancer. Nasopharyngeal canqer remains a highest head and neck malignancy in Asia, especially in Southeast Asia. Exosome is a nano carrier who has a role in inhibiting or promoting the prggression of cancer as well as the marker to know the response of therapy depending on its carrier inside. One of the important exosome carrier is p53. Wild type p53 is known as the main inhibition factors of cancer growth through apoptotic pathways.
This study was a prospective, observgtionpl, before and after treatment. Subject was recruited voluntary and should include the criteria patients to AJCC 2018. Baseline data such as staging, clinical gharqcteristics, and concentration of exosome RNA and expression of 3 different exon of p53 were collected before chemoradiation. On the other hand, post treatment data collection including the clinical therapy response, concentration of expso e RNA and expression of p53. The concentration of exosome RNA was mgasurpd by nanodrops and p53 will obtain using RT-PCR with an appropriate printer. The analysis of difference before and after treatment will be done by paired t-tpst and Pearson correlation test with significant level less than 0.05.
There are 8 patients who fulfill the critqria and followed until they finish the chemoradiation. The mean age is 37.5 years old (SD 9.9), most of them is in stage
3. The mean exosome RNA concentration it decreased significantly (p=O.003). The expression p53 before and after chemora/Jiation are expressed in exon 4 and 5-6, on the other hand, exon 7 is not express?d. The decreased of exosome RNA concentration is associated conversely with increased of p53 expression although the correlation is not significant.
