Abstract (Abstrak)

ASTRI ALFRIANTI. Growth Ability of Lactic Acid Bacteria Isolates from Free- Range Chickens Gallus domesticus and Female Ducks Anas domesticus Using Skim Milk as a Coating Agent, (supervised by Prof. Dr. Dirayah Rauf Husain, DEA). Baground. Lactic acid bacteria (LAB) play a crucial role in the food and health sectors due to their ability to produce bioactive metabolites and are excellent probiotic candidates. However, maintaining their viability and stability, especially for long-term use, requires appropriate conservation methods. Objectives. Analyzing the growth ability and generation time of LAB isolates from native chickens (PATA-5) and female ducks (PRIB-3), after conservation using skim milk coating material. Methods. This study consisted of 5 stages, namely: 1) Characterization of LAB isolates through purity tests, morphological observation, physiological tests, and biochemical tests; 2) Biomass production based on growth curves; 3) Inhibitory test againts pathogenic bacteria; 4) Conservation of LAB isolates through coating using skim milk, lyophilization for 49 hours, and storage at room temperature for one week; 5) Viability test. Results. LAB isolates PRIB-3 and PATA-5 showed good growth ability after conservation, with optimal biomass increase at 6 to 10,5 hours. Inhibition test showed that the inhibition zone of PRIB-3 lyophilisate ranged from 9-13.5 mm, while PATA-5 ranged from 10-12 mm. Both isolates were classified as having strong to very strong inhibitory activity. The generation time of PRIB-3 was recorded at 4 hours 30 minutes, while PATA-5 was 4 hours 15 minutes, which is still within the normal range for LAB and not significantly different compared to before conservation. Conclusion. LAB isolates PRIB-3 and PATA-5 had good growth ability after conservation using skim milk as a coating material.
