Abstract (Abstrak)
ABSTRACT NURQALBI SULFA. Intact Plasma Membrane and Intact Acrosome Cap of Saanen Goat Spermatozoa After Sexing Using Freeze-Dried Albumin with Different Incubation Times (supervised by Muhammad Yusuf). Spermatozoa sexing technology is one of the methods used to improve livestock reproductive efficiency through controlling the sex of offspring. In Saanen goats, which are dairy animals, the birth of female offspring is preferred due to their potential for milk production. The use of freeze-dried albumin as a sexing medium has been shown to facilitate the separation of X- and Y-bearing spermatozoa; however, the incubation period may affect sperm quality. This study aimed to determine the effect of different incubation times on the integrity of the Plasma Membrane (PMI) and Acrosome Cap (ACI) of sexed Saanen goat spermatozoa using freeze-dried albumin. The study was conducted from March to April 2026 at the Animal Reproduction Laboratory and Animal Center, Faculty of Animal Science, Hasanuddin University. A Completely Randomized Design (CRD) was employed with four treatments: P0 (control), P1 (30 minutes), P2 (60 minutes), and P3 (90 minutes), each with four replications. The observed parameters included Plasma Membrane Integrity (PMI) and Acrosome Cap Integrity (ACI) of spermatozoa. Data were analyzed using Analysis of Variance (ANOVA) followed by Duncan’s Multiple Range Test. The results showed that different incubation times had a significant effect (P<0.05) on the PMI and ACI of Saanen goat spermatozoa. The highest PMI and ACI values were obtained in treatment P1 (30 minutes), while the lowest values were observed in treatment P3 (90 minutes). Longer incubation periods tended to decrease sperm quality due to damage to the plasma membrane and acrosome cap during the sexing process. Based on the results of this study, an incubation time of 30 minutes was the most optimal treatment for maintaining the quality of sexed Saanen goat spermatozoa using freeze-dried albumin.
