Abstract (Abstrak)
Omphalitis merupakan salah satu penyakit penting pada day-old chick (DOC) yang ditandai oleh infeksi pada umbilikus dan yolk sac serta menjadi penyebab utama peningkatan mortalitas pada minggu pertama kehidupan. Penelitian ini bertujuan untuk mengidentifikasi tanda klinis, perubahan patologi anatomi, serta bakteri yang diduga berperan pada kasus omphalitis pada DOC di hatchery PT New Hope Indonesia. Pemeriksaan dilakukan terhadap empat ekor DOC berumur 1–5 hari yang menunjukkan gejala klinis mengarah pada omphalitis. Metode yang digunakan meliputi anamnesis, pemeriksaan fisik, nekropsi, isolasi bakteri menggunakan media Eosin Methylene Blue Agar (EMBA), dan pewarnaan Gram. Hasil pemeriksaan menunjukkan DOC mengalami kelemahan, bulu kusam, umbilikus yang tidak menutup sempurna, distensi abdomen, dan pembesaran yolk sac. Hasil nekropsi menunjukkan adanya retensi yolk sac, peritonitis fibrinopurulen, adhesi antara kantung kuning telur dengan organ viseral, serta hemoragi pada paru dan otak. Pemeriksaan bakteriologis pada media EMBA menunjukkan pertumbuhan koloni berwarna gelap dengan kilau hijau metalik, sedangkan hasil pewarnaan Gram menunjukkan bakteri berbentuk batang Gram negatif. Berdasarkan hasil pemeriksaan, kasus omphalitis pada DOC diduga disebabkan oleh infeksi bakteri Gram negatif, terutama Escherichia coli, yang berpotensi berkembang menjadi septikemia sistemik. Faktor predisposisi yang diduga berperan meliputi sanitasi hatchery yang kurang optimal, fluktuasi suhu dan kelembapan inkubator, serta penyimpanan telur tetas yang tidak sesuai prosedur. Omphalitis is a major disease in day-old chicks (DOCs), characterized by infection of the umbilicus and yolk sac and a major cause of increased mortality in the first week of life. This study aimed to identify clinical signs, anatomical pathological changes, and bacteria suspected of contributing to omphalitis in DOCs at the PT New Hope Indonesia hatchery. Four DOCs aged 1–5 days exhibiting clinical signs suggestive of omphalitis were examined. The methods used included history taking, physical examination, necropsy, bacterial isolation using Eosin Methylene Blue Agar (EMBA) media, and Gram staining. The examination results showed that the DOCs experienced weakness, dull feathers, an incompletely closed umbilicus, abdominal distension, and an enlarged yolk sac. Necropsy results revealed yolk sac retention, fibrinopurulent peritonitis, adhesions between the yolk sac and visceral organs, and hemorrhage in the lungs and brain. Bacteriological examination of EMBA media showed dark colonies with a metallic green sheen, while Gram staining revealed Gram-negative rod-shaped bacteria. Based on the examination results, the omphalitis cases in DOCs are suspected to be caused by Gram-negative bacterial infections, particularly Escherichia coli, which have the potential to develop into systemic septicemia. Predisposing factors suspected to play a role include suboptimal hatchery sanitation, fluctuations in incubator temperature and humidity, and improper storage of hatching eggs.
