Abstract (Abstrak)

Sarniati R.R. In Vitro Study on The Inhibition of α -Amylase and α -Glucosidase
Activity by Kleinhovia Hospita Linn Leaf Extract And Fractions. (Supervised by
Yulia Yusrini Djabir and Abdul Rahim)
Kleinhovia hospita is a herbal plant that has long been used to treat
various diseases, one of which is diabetes mellitus. This study aims to evaluate
the inhibitory effect of α-amylase and α-glycosidase enzyme activity by K.
hospita ethanolic extract and its fractions. K. hospita leaves were extracted with
96% ethanol then liquid-liquid partitioning was carried out using n-hexane, ethyl
acetate, and water as solvents. The inhibition of the ethanolic extract and its
fractions on the activity of the α-amylase and α-glycosidase was determined
using an enzymatic reaction that was measured spectrophotometrically at 540
nm and 405 nm, respectively, to obtain % inhibition based on the linear
equation and the IC50 value. Acarbose was used as the positive control.. The
results showed that the IC50 values of the ethanol extract, n-hexane fraction,
ethyl acetate fraction, and water fraction were 1496.8 µg/mL, 1266.7 µg/mL,
12577.5 µg/mL, and 5217.0 µg/mL, respectively, against α-amylase activity. As
for α-glucosidase activity, the IC50 values were 1341.2 µg/mL, 25470.0 µg/mL, 64450.0 µg/mL, and 1291.4 µg/mL, respectively. These values were far greater than acarbose, namely 7.4 µg/mL for α-amylase activity and 5.5 µg/mL for α-glucosidase activity. The high IC50 values of the extracts and fractions of K. hospita indicated that K. hospita did not have an anti-diabetic effect through the activity of inhibiting the α-glucosidase and α-amylase
