Abstract (Abstrak)

Faigah Aprilia. Analysis of Vascular Endothelial Growth Factor in Platelet Rich Plasma with Addition of Activator Calcium Chloride and Thrombine in Different Temperature and Time of Storage (supervised by Uleng Bahrun, Tenri Esa).
Background: Platelet rich plasma (PRP) can potentially increase the healing process through growth factor (GF) and α-granular cytokines contained in platelets. Vascular endhotelial growth factor (VEGF) is one of the largest GFs in PRP with a strong ability to induce endothelial cell permeability, proliferation, angiogenesis which has an important and applicable role in various fields of medicine. The aim of this study was to compare the platelet count and VEGF PRP levels with the addition of calcium chloride (CaCl2) and thrombine as activators at different temperature and time of storage.
Methods: An experimental laboratory design using 16 samples was perfomed by measuring VEGF PRP levels by ELISA method before activation (VEGF PRP) as the initial examination. Platelets then activated using CaCl2+thrombine activator at 40C for 30 minutes, and VEGF PRP levels measured after activation for 1 hour at room temperature (t-PRP1), 24 hours at 40C (t-PRP2) and 7 days at -200C (t-PRP3). Data were analyzed using the Wilcoxon, Friedman and post hox Wilcoxon tests, significant if p<0.05.
Results: There was a median VEGF increase of 1.07 times after 1 hour of activation at room temperature (p=0.049), a decrease of 0.88 times after 24 hours of activation at 40C (p=0.642), an increase of 1.83 times after 7 days of temperature activation -200C (p<0.001). It was concluded that PRP activation with CaCl2+thrombin for 7 days at -200C produced the highest VEGF compared to room temperature and 40C. It is recommended to avoid using PRP stored at 24 hours at 40C.
