Abstract (Abstrak)

Inflamasi vaskular merupakan salah satu faktor penyebab terjadinya patogenesis penuaan vaskular. Aktivasi faktor nuklear-κB (NF-κB) akan menginduksi molekul-molekul adhesi seperti VCAM-1 dan ICAM-1 yang akan menyebabkan migrasi dan adhesi sel monositik ke sel endotel vaskular, yang berperan penting selama proses inflamasi vaskular. Trehalosa merupakan non-reducing disakarida yang memiliki beberapa fungsi yaitu protektif terhadap stressor (salah satunya ROS) dan mencegah respon inflamasi yang diinduksi oleh syok endotoksik. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian gula trehalosa terhadap proses inflamasi makrovaskular terkait proses penuaan. Penelitian ini menggunakan hewan coba tikus Wistar (Rattus novergicus) jantan sebanyak 28 ekor yang dibagi menjadi 4 kelompok yaitu kelompok kontrol tikus muda (Kelompok A), kelompok kontrol tikus tua (Kelompok B), kelompok tikus tua yang diberikan larutan sukrosa (Kelompok C), dan kelompok tikus tua yang diberikan larutan trehalosa (Kelompok D); yang kemudian diamati selama 8 minggu. Pemeriksaan imunohistokimia (IHC) ekspresi NFκB-p65 jaringan aorta digunakan untuk menilai proses inflamasi makrovaskular pada penelitian ini. Hasil penelitian menunjukkan tidak ada perbedaan yang signifikan antara ekspresi NFκB-p65 jaringan aorta pada kelompok kontrol tikus muda dan kontrol tikus tua (p=0,247). Kelompok trehalosa memiliki ekspresi NFkB-p65 jaringan aorta lebih rendah dibandingkan dengan kelompok kontrol tua (p=0,012); sedangkan pada kelompok yang diberikan larutan sukrosa memiliki ekspresi NFkB-p65 jaringan aorta lebih tinggi dibandingkan dengan kelompok trehalosa (p=0,018).
Kesimpulan: Trehalosa memiliki efek positif terhadap proses inflamasi vaskular terkait penuaan yang dapat dilihat dari rendahnya ekspresi NFκB-p65 jaringan aorta pada tikus tua.

Vascular inflammation is one of contributing factor for the pathogenesis of arterial ageing. Activation of nuclear factor-κB (NF-κB) will induce adhesion molecules such as VCAM-1 and ICAM-1 which will cause migration and adhesion of monocytic cells to vascular endothelial cells, which play an important role during vascular inflammation processes. Trehalose is a non-reducing disaccharide that has several functions, protecting against stressors (one of them is reactive oxygen species/ROS) and preventing the inflammatory responses induced by endotoxic shock. The purpose of this study was to determine the effect of trehalose sugar supplementation on macrovascular inflammatory processes related to the aging process. This study used 28 male Wistar rats (Rattus novergicus) as experimental animals which were divided into 4 groups, control group of young rats (Group A), control group of old rats (Group B), group of old rats that given sucrose solution (Group C), and group of old rats that given trehalose solution (Group D); were then observed for 8 weeks. Immunohistochemical examination (IHC) of aortic tissue NFκB-p65 expression was used to assess the macrovascular inflamsmatory process in this study. The results showed that there were no significant differences in aortic tissue NFκB-p65 expression between old and young subjects (p=0,247). The trehalose group had lower aortic tissue NFkB-p65 expression compared to the old control group (p=0,012); while the group given sucrose solution had a higher aortic tissue NFkB-p65 expression compared to the trehalose group (p=0,018).
Conclusion: Trehalose has a positive effect on aging-associated vascular inflammatory processes that can be seen from the low aortic tissue NFκB-p65 expression in old rats.
